Gene expression in CA3

A sensitive and efficient cognitive screening assay for use in model development and target validation.

Current mouse behavioral paradigms are time-consuming, and lack both sensitivity and selectivity, as they are subject to numerous confounding variables and multiple neural pathways may contribute to changes in performance. These tests thus often produce false negative or positive results, and do little to indicate a mechanism of action.

Solution

A modified environmental exploration protocol has been designed such that performance is equivalent across subjects and experimental groups. Under control conditions, this protocol engages cortical and hippocampal neurons responsible for spatial and working memory, as well as multiple sensory pathways. Subsequent measurement of the extent of activation provides a window into the ability of involved neural systems to encode the new information.

We offer two design levels:

  1. Screening Study Design offering rapid measurements of induced gene expression within brain regions.
  2. Mechanism Validation Design with simultaneous measurement of pathology and activation in individual neurons.

This protocol has currently been validated in several Alzheimer's disease models and is broadly applicable to study of neurodegenerative disease.
